If you are applying for a position do you inform your supervisor already or you have to wait if you are already shortlisted in order not to pre-empt?

This depends very much on your specific working relationship with your supervisor, if you have already communicated your wish to advance and take on new and more challenging positions in the future as part of your SES this may not come as a big surprise. Keep in mind that when you apply for a position in many cases you will be asked to provide the name of your supervisor.
